    Activités antioxydantes, antibactériennes et anti-α-amylases de divers extraits d'Anvillea radiata récoltés dans la région de Ghardaïa

    Current research is focused on molecules endowed with biological activities of natural origin. The objective of this study is to evaluate the antioxidant activity, the antibacterial and anti-α-amylase activities of various extracts of Anvillea radiata aerial part the (Asteraceae) from the region of Ghardaia, which is widely used in traditional medicine in Algeria. The extraction of the active ingredients of Anvillea radiata aerial part was carried out by cold maceration using three solvents of different polarities (methanol, ethyl acetate and petroleum ether). Phytochemical analysis of the various extracts is performed by phytochemical screening and quantification of some phenolic compounds classes. Several biological activities were evaluated namely the antioxidant activity using three tests (ABTS, FRAP and bleaching of β-carotene), the antibacterial activity by diffusion on agar medium and by microdilution method and the antidiabetic activity by the measurement of α-amylase inhibition. The obtained results showed the richness of the studied species in phenolic compounds (tannins, flavonoids, coumarines ... etc.) and terpene (saponosides and steroids). The highest contents of total polyphenols, flavonoids, phenolic acids and condensed tannins are obtained with the methanolic extract which are in the order of 81.38 ± 19.16 mg EAG / g, 16.37 ± 0 , 89 mg ER / g, 4542.00 ± 8.49 μg EAC / g and 33.97 ± 1.06 mg EC / g respectively. The methanolic extract of Anvillea radiata has the best inhibitory capacity of the ABTS •+ radical cation , iron reducing power and β-carotene bleaching inhibitory effect with IC50 equal to 153.48±10.02 μg/ml, 95.88±9.13 μg/ml and 172.94±14.88 μg/ml respectively comparable to those of the standard antioxidants used. As regard to the antibacterial activity, the ethyl acetate extract has the potential capacity to inhibit bacterial growth. on both strains tested (S. aureus and E. coli) with MIC equal to 0.33 mg/ml. The best inhibitory activity of α- amylase was recorded with the methanolic extract where the IC50 value of 76.31±5.60 μg/ml, which is lower than that of acarbose. In conclusion, the obtained results may justify the traditional use of this species to treat inflammatory problems, infectious diseases and diabetes.

    Dosage des polyphénols et des flavonoïdes d’Ammodaucus leucotrichus et Cotula cinerea

    Résumé Les plantes étudiées Ammodaucus leucotrichus et Cotula cinerea ont été récoltées et conservées par séchage, les échantillons obtenues ont été analysés pour déterminer leurs rendements et leurs teneurs en PPt et Ft, l’extraction est réalisée par macération avec quatre types de solvants de polarités différentes. Les résultats obtenus montrent que le rendement le plus élevé pour Ammodaucus leucotrichus est obtenu avec l’acétone (4.16%) alors que pour Cotula cinerea le rendement maximale est enregistré avec l’extrait aqueux (2.66%). La teneur en polyphénols des différents extraits est déterminée par la méthode de Folin-Ciocalteau. Nous constatons que les deux espèces présentent des teneurs en PPt très proches pour chaque type d’extrait. La meilleure valeur de taux des PPt est obtenue avec les extraits aqueux pour les deux espèces qui sont de 54364.58 ±1398.01μg EAG/g pour Ammodaucus leucotrichus et 52812.50 ±891.04 μg EAG/g pour Cotula cinerea. Nous avons aussi évalué la teneur en flavonoïdes des différents extraits par la méthode de KIM et al. (2003). Les deux espèces Ammodaucus leucotrichus et Cotula cinerea présentent des teneurs en flavonoïdes les plus élevées pour les extraits aqueux et qui sont 921.21 ±73.48μg et 854.54 ± 20.99 μgER/g du poids sec respectivement. Il ressort de ces résultats que les teneurs en PPt et Ft sont proportionnelle à la polarité du solvant d’extraction pour les deux plantes ce qui montre que ces espèces sont riches en composés phénoliques glycosylé. ملخص تم جمع النباتات التي تمت دراستها: Ammodaucus leucotrichus وCotula cinerea تم حفظها بالتجفيف، وتم تحليل العينات التي تم الحصول عليها لتحديد إنتاجيتها ومحتوياتها من PPt وFt، يتم إجراء الاستخلاص عن طريق النقع بأربعة أنواع من المذيبات. أقطاب مختلفة. أظهرت النتائج التي تم الحصول عليها أن أعلى عائد ل يتم الحصول على Ammodaucus leucotrichus باستخدام الأسيتون (4.16٪) بينما يتم الحصول على Cotula cinerea يتم تسجيل الحد الأقصى للإنتاجية باستخدام المستخلص المائي (2.66%). محتوى البوليفينول يتم تحديد المستخلصات المختلفة بواسطة طريقة Folin-Ciocalteau. نلاحظ أن كلا تحتوي الأنواع على محتويات PPt متشابهة جدًا لكل نوع من أنواع المستخلصات. أفضل قيمة تم الحصول على معدل PPt للمستخلصات المائية للنوعين وهي 54364.58 ±1398.01μg EAG/g لـ Ammodaucus leucotrichus و52812.50 ±891.04 ميكروغرام EAG/g لـ Cotula سينمائي. قمنا أيضًا بتقييم محتوى الفلافونويد في المستخلصات المختلفة بطريقة كيم وآخرون. Abstract The studied plants Ammodaucus leucotrichus and Cotula cinerea were harvested and preserved by drying, the obtained samples were analyzed to determine their yields and their PPt and Ft content, the extraction is realized by soaking with four types of solvants of different polarities. The obtained results show that the the highest yield, for Ammodaucus leucotrichus is obtained with the acetone (4.16%) while for Cotula cinerea the maximum yield is recorded with the aqueous extract (2.66%). The polyphénols content of the different extracts is determined by the folin- ciocalteau method. We note that the two species present PPt content very close for every type of extract. The better value of rate of the PPt is obtained with the aqueous extracts for the two species that are of 54364,58 ±1398.01μg EAG/g for Ammodaucus leucotrichus and 52812,50 ±891.04 μg EAG/g for Cotula cinerea. We also evaluated the flavonoïdes content of the different extracts by the method of KIM and Al. (2003). The two species Ammodaucus leucotrichus and Cotula cinerea present flavonoïdes content the more students for the aqueous extracts and that are 921,21 ±73.48μg and 854,54 ± 20.99 μg ER/g of the dry weight. Respectively it appears of these results that the PPt and Ft content are proportional to the polarity of the solvant of extraction for the two plants it self that shows that these species are rich in phenolic compounds glycosylated.
